I have a 250 GB SSD drive that has my Win 7 operating system and a few critcal programs loaded on it (Lightroom, Photoshop etc...). The rest of my programs and data are stored on a 2 GB 7200 rpm drive.
Question: Should I store the LR4 catalog on the C: Drive (SSD), but store all of the previews, back-ups and other folders on the D: Drive?
What is the best way to install the program and place all of the folders in a multi drive system?
Ad 1.:
Yes, the previews need good reading speeds, too.
Ad2.:
Adobe wants the previews folder next to the lrcat-file.
You can deviate from that using a symbolic link for an alternative location for your previews folder.
Victoria Bampton describes this in her Missing FAQs LR4:

Yes, placing the catalog on the SSD will help. However, as you've noted the previews folder, especially with 1:1 previews, will likely require more space than you have avaialble. You can, however, relocate the previews folder/package to another drive, but the link to it must be made to it using a symbolic link; shortcuts and aliaes don't work. See this post for more info on how to go about it http://forums.adobe.com/thread/778350
