I made a copy of a layer (a colored photo that I downloaded from Bridge) and I was messing around with the adjustment settings like desaturation. However, when I clicked on the adjustments tab most of the selections were greyed out. Do I need to go up to color settings and change them to fix the problem?

thauser79, is a layer mask targeted in the layers panel? A screenshot would help, as would some basic info about your setup, the version of Photoshop that you are using.
You seem to be asking a lot of basic questions, did you take some time to peruse the user guide? http://helpx.adobe.com/photoshop.html (a book is useful too, otherwise, you'll hit a lot of frustrating stops if you do not learn the basics first)
Christoph, I think that Doug meant that most filters work only with 8bits files.

Yes this is an overstatement by me....I meant image tools...many image and color operations only work with 8 bit images not 16 bit....but lots of other tools still work
For example can't save a 16 bit image to .jpg (file format limitation, not a Ps limitation per se) or applying many filters
check image bit setting: menu bar, image, mode: _____
