In a german forum, someone found a bug about having hyperlinks in a multi-column layout. All Hyperlinks starting the second column do not work. In between more people (me included) can confirm this bug in v20 and v21. You have to create single frames to get it work.
Please Adobe representatives, forward this bug report to the engineers.
As I already wrote on that German forum, I can confirm that bug for InDesign CS5.5 and v20 DPS. Did not test in newer version.
What perplexed me is, that if I go to the Adobe "Feature Request/Bug Report Form" site there is no way to address a bug directly to DPS or a specific DPS version:

I did the tests today with CS5.5 and CS6, a v20 folio and a v21 folio. Always the same result/bug.

I mentionned it here in the forum as I do not know if and and how reliable the bug report form will be read by the responsible staff at Adobe.
This is the probably the most monitored of the user forums by Adobe personnel, but I would still file the bug report. I very rarely use hyperlinks and opt for buttons instead. I've found hyperlinks a bit wonky.
BTW, one thing to try...do not use shared hyperlink destinations.
