What are some reasons I would be unable to use the Paint Brush tool to pain on a layer?
When I try to paint it says the layer needs to be rasterized first. But even when I click ok and start to paint it's not painting black which is the color I set the brush to paint. It's more like it's erasing but not quite.
The layer doesn't have a mask or anything.

Considering your "message," I think that Noel has narrowed down the exact problem that you are facing - you are trying to Paint onto a Vector Layer.
Something else, that can affect the ability to Paint (or employee many other functions) is if you have a Selection (perhaps one that is Hidden), and are trying to Paint outside of that Selection. There, however, you will not get the message, that you mention. Just something else to think about, when Painting is not working.

No, rasterizing the layer didn't help. When I did click the raster option and would paint the color was muted and seemed more like it was erasing.

Well, sorry to return days later but I developed a head cold shortly after posting this question. Still fuzzy in the head so I'm not quite able to concentrate on a better response.
