When purchasing the trial version of CS6 does it have to be re-downloaded a second time?

No. Once you purchase the product convert, you can convert the product to a licensed version, by entering the serial number or even to a subscription. You do not need to download again. See http://www.adobe.com/uk/special/products/creativesuite/trial/faq/index .html
