This seems to be happening to large files in general, but happens more often when saving down from CS6 to CS3 (for preflighting). The files will save, then throw this error when re-opening. The preview of the file in the Finder will disappear and the file becomes useless. We are on Macs running 10.6.8 and Illustrator CS6 (fully patched).

In some cases you may be able to open a new document and then File>Place the damaged file.
Another way, sometimes possible, is far more complicated and involves mending the code. The following Illustrator Help pages describe part of it. In the case of parts missing, it is necessary to add parts of the damaged file into a sound one, if sufficient viable parts are present.

Obviously your issue should nor occur, and certainly not regularly.
Earlier, it has seemed that file corruption has often/mostly been a matter of errors in connexion with saving, caused by the OS.
Obviously, this may have changed with the newest versions, and maybe especially with downsaving.
It has not been up here before, I believe.
