You don't have to change its value; just leave it at 100%. It also doesn't matter what frame you use to set the keyframe.
This should be a sticky or a FAQ entry.

Older versions of Pr had a default opacity keyframe set automatically, which is counter-intuitive to most motion graphics artists and video editors who have experience with animation. Typically, editors won't want *any* animation keyframes set unless they do it themselves. Just look at After Effects.
CS4 doesn't have the default keyframe, and I suspect that is the source of this issue.

Ann Bens Dec 28, 2008 7:34 AM (in response to (Kees_Jansen))I am not a motion graphics artist but this way is much beter, being able to set your own keyframe instead of Premiere doing it for you. I was always busy removing them.
But now that the new 'old' way messes up the transitions is indead a bug. -
