How to create a selection from a shape on a transparency?

I've been watching Corey Barker's video "Adobe Photoshop: Starting from Scratch." Great stuff, except he performs the same operation in two different segments without explaining what he did -- a real show-stopper for a newbie like me.

Ex. "Lights On" at 3:07. Here he takes a transparency layer with a shape on it and creates a selection out of that shape/object, from which he later creates a layer mask on another layer.

I'm sure it's simple, but exactly how do you create a selection out of the shape of an object that's on a transparency? Thanks.
