Perhaps the effects enable toggle was turned off?
If that switch is disabled, all parameters for an effect are subsequently disabled and not passed through to the final composite. It's basically just an on/off switch for the effect; you can change the parameters all the live long day, but they won't have any effect.

Thanks for your replies. I have now deleted the clip from the timeline and re-added. All is ok, but i dont know why. The effect switch was on
