In Illustrator 15 Save for Web shows overprints…
Or you can use Flatten Transparency. If all you have are overprints and all objects have 100% Normal opacity then you need to force flattening. Draw a white rectangle over everything and set it to Multiply.

Actually What I want to do is that there is my original document containing some arts or objects like as follows:
Currently doc is set to CMYK/Preview. Now I clicked the menu item Window>Separations Preview where i checked the Overprint preview, this will set my document CMYK/Overprint Preview and I unchecked Cyan, Yellow and Black from separation preview dialog which gave me result like this :
I am trying to export the above overprint preview but every time I got the original one.
So I am just curious can it be doable or not?
CS 6 also shows thee preview in save for the web and in Acrobat I believe if you use output preview in the productions tools you see see the preview as well but only while the output preview window is opened.
Old post, but if anybody is trying to do this, you can install a image printer, an virtual print that print your files in jpg, tiff or other file type. Than, in print dialog box, you can set illustrator to simulate overprint.
In the latest Illustrator version you can just export as png or jpeg and the Overprint will be shown in the resulting file.
