Can I transfer my Digital Edition Library to a flash drive, and external hard drive or SD card and the remove it from my hard drive, (need to clean up my hard drive and make some room) and still be able to transfer books to my Nook or other devices.

You should be able to.  Make sure the Nook etc are registered with the same AdobeID as Digital Editions.

Then you should be able to move DRM .epub files between the two;

directly (Windows Explorer etc), or via SD cards, email (depending if it is a tablet or simple eReader), etc.

Once the file finally arrives on the Nook, it should still be readable.

But try it out before you commit yourself too much; the DRM system always seems to find new nasty surprises.....
