Assuming you use DirectLink, any grades you make in SpeedGrade (even if they are on different grading layers) get combined into a single Lumetri effect on that clip when you DirectLink back to Premiere Pro.
If you edited in Premiere Pro with the RED 4K footage then DirectLink is probably the easiest way to go. However if you edited with low-res offline transcodes of that RED footage, an EDL is probably your best bet.

To have separate grading layers in Sg in a PPro->Sg direct-link workflow you MUST create adjustment layers in PPro before sending the sequence to Sg. Sg is NOT allowed to mess with the timeline of a PPro project is the reason for this.
So ... starting in Premiere Pro ... I'll typically create an adjustment layer over an individual tough clip or one that may need extra stuff of masking et al; I may create a multi-clip adjustment layer over a group of very similar files, so I'll end up with a way to adjust them to the footage around them as a group; and I may make an over-all grading layer if I am going to apply some effect to the whole shebang.
Direct-link to Sg, do a primary or first-pass grading on the footage of the clips themselves, copying some work to other clips in the timeline, using the layers palette in Sg. Then work on the adjustment/grading layers (from PPro) for special needs, then save/back to PPro.
Works pretty nice really.
