Full color RGB, 256 Grey degree scales, whats that all about?!

Can anyone help? I want to start drawing images for an LED matrix sign. The manufacturers state that the sign is full colour RGB, 256 grey degree scales. What is the best programme i.e photoshop, indesign or illustrator to begin drawing these images and how should I set up the template to achieve the best colours?

I can see that the settings on photo shop have the option to set up as RGB or Greyscale and various 'bits'. This is all going over my head!
