Brush animation based on video

I'm going to preface this by saying that I am a beginner.

I have been drawing over a video with the brush tool to make an animated effect on it. I would like to somehow isolate the brush strokes from the video, so instead of having brush on top of video I could make it be brush on top of some different background. Is it possible to do this, and how..?

I've looked up how to do this, but I haven't found any answers. If anyone has advice, please let me know!

check the paint on transparent checkbox

if this does not solve your problem, show some screenshots and be more specific as to what is not working
