Start by rendering to a different location. I mean Desktop? RLY? You can easily run out of space on that one since it has a built-in quota and even if that weren't an issue, Windows keeps checking and updating your desktop regularly, so interfering file operations can easily mess up a render in progress when it tries to generate a thumbnail/ preview.

Can you render your AE composition with the AE render queue into an intermediate codec (like DNxHD, Cineform, or QuickTime with the Photo-JPEG codec)?
