It could very easily be your system isn't up to snuff. Hard to tell without details.
I doubt that is it, it's a brand new msi gt70, core i7 12GB of RAM, GDDR5 3GB Graphics Card
Can anyone help with this? I tried re-downloading premiere pro with the same results.

HDD: 750GB, but I run off of an external 3TB drive
When I scrub through the timeline it shows everything in the playback panel, but doesn't work when I play it. I have CS5.5 on another laptop that is the exact same and havent had any problems whatsoever. I also find it hard to believe that the system isn't up to snuff because I just rendered 3 400 shot RAW timelapses shot on a full frame Nikon D600 in After Effects with no problems.
Presumably Premiere Pro is installed on the internal system drive, and your media is on the external 3GB drive?
How is the external drive connected to your laptop? USB2, USB3, eSATA?????
What is the format and source of your video clips?
Meida only is on the 3TB drive

I found that the Nvidia graphics card was not being used hence the problem. After disabling the intel card - no problems
