Welcome to Adobe Community.
This could be because of corrupt preferences.
We could try to recreate the preferences, however it will delete your customizations in the workspace. But we will not completely delete the preferences folder so we can get it back.
In order to recreate the preferences, please do the following.
Save your Project.
Close Captivate
Windows 7: C:\Users\[user name]\AppData (hidden folder)\Local\Adobe\Adobe Captivate 6 folder
Windows XP: C:\Documents and Settings\[user name]\Local Settings\Application Data\Adobe\Adobe Captivate 6 folder
Mac: /Users/[user name]/Library/Preferences/Adobe Captivate 6 folder
Rename the Captivate 6 folder to anything.
Launch Captivate and test your project.

When this happens to you next time, perhaps it would be helpful to double check a place you may have clicked and not realized it. Sure, clobbering the preferences nailed it, but that's a bit like using a bazooka to kill the fly in the room.
Too late now, but my guess is that this is what happened.
If you look at the Timeline area you should see a small speaker icon as shown below:
It's easy to click this and there isn't much of an indication you have muted the slide. My guess is that you managed to click this earlier. Too bad you had to clobber preferences to get it back.

This fix is not working for me.
Info:
I am running 6.0.1.240
I am recording as software simulation (I did try video demo and i can't hear that either)
I have Admin Access and I make sure to use it when running the application
I have checked the little speaker button and it is on.
I have deleted the preferences.
I also tried reloading the software.
Note: When I play audio in edit mode, all I get is silence. If I play the slide in the Web Browser, I CAN hear my audio, so I know it is recording.
