I am trying to roto a 1:05 clip with roto brush in AE CC. When I get 34 seconds in and click to the next frame the mask disappears. If I go back one frame it’s there then forward again and it disappears as if there is no roto. I have 32 gig ram and a large cache. What am I doing wrong? How do I roto one person out of a clip that long?

You are probably creating new base keyframes because you are not allowing the propagation to do its magic. Provide screenshots to clarify.
Make sure you know how to use rotobrush. Here are a bunch of free resounces from searching After Effects Help inside AE:
You may not have set the range for auto propegation corectly.
There is also no reason you can't split the clip. In most cases folks roto way more than they need to and roto way too much detail. The average shot in a movie is less than six seconds.
By 1:05, do you mean 1 minute and 5 seconds? That's a really long take. Have you edited this clip?
