Which version of Flash, which platform? Do you have any other apps running? Does something unexpected happen when you use the keyboard shortcut, or does nothing happen?

Flash CC, Win7 64bit, I use sublime text, nothing else is running...
Absolutly nothing is happening if I try.
Correction: ctrl+enter has been successfully assigned (I see the text in the menu) and is recongnize for "test movie ...air debug desktop"
Others don't appear in the sub-sub-menu for testing or debugging over usb.
I've assigned them to ctrl+alt+enter/return and ctrl-shift-enter/return. (I've tried other keys as well...)
So it appear the problem is with test/debug USB...
EDIT: Just to be clear, I try to set custom shortcuts, to avoid the default behaviour (one shortcut to launch the last choice). this weird behaviour is probably the cause of the problem in the first place.

If the built-in keyboard shortcuts work and your custom shortcuts don't work, then I'm guessing that the problem is with the custom shortcuts. You could remove all of your custom shortcuts, then set one new shortcut and see if that one works. If it does, then try and build on that by adding additional shortcuts, one at a time.
Well, all other custom shortcuts were working.... but I just did another set from the default one, assigned one key that I knew was working, and still nothing.
I think its a genuine bug;

If I remember correctly I think it didn't worked either with CS6...
If you search around these forums you'll see no shortage of key binding issues of all sorts. Chances are you're finding another. It'd be useful to report it on the bug report form. The more bugs rolling in surrounding keyboard mapping the more they'll take it serious in the next update:
I can verify I cannot set a working shortcut for this in CC, but it works fine in CS6.
