Manually reset the permissions on your user/Library/Preferences/Adobe/After Effects folder and grant permission to all except everyont to read write, then apply to enclosed folders. Reboot and try opening again.

Tried this fix but it still doesn't work. Still hanging on "Initializing MediaCore"
I should add that Activity Monitor shows that both After Effects, as well as Adobe QT32 Server, are not responding. I've tried force-quitting the Adobe QT32 Server to see if that was stalling out AE, but with no effect.
OK... trashed the preferences folder and now it is working. Go figure.

I am experiencing the same thing after updating to 12.2 on the PC side. Anyone?
Here's the crazy thing with AE failing while "Initializing MediaCore": both Media Encoder CC and Premiere Pro CC both open and operate completely. This only happened after upgrading to 12.2.
Since then, I have uninstalled 12.2, and following your advice outlined in the link above, I let CC Manager install AE 12.2 to the system drive (the rest of the suite is installed on a program drive [E:] because my SSD system drive is only 128GB). It still fails to launch. I will now uninstall again, but this time, I will remove all Preferences with the Uninstall / Control Panel option which affects the DynamicLinkManager.exe, Adobe Media Encoder.exe, and Adobe QT32 Server.exe....worried!
I'll let you know what happens.
Well, I re-installed to a new drive, and that didn't help. I then donwloaded the Adobe Cleaner utility, ran it and removed all traces of After Effects on my system but keeping the preferences, re-ran the CC Manager installation of AE. After the installation, I ran the program, and it failed to launch while initializing MediaCore. I then ran the Cleaner utility, this time removing all of AE and preferences. Re-installed AE, and it sucessfully opened the program only to close 1 second later with no indication why it closed. I then ran the Cleaner utility, this time removing ALL Adobe software. Re-installed CC Manager, ran the installer for AE, completed the installation, launched the program, and WHAMMO, it closed after 1 second!
I am dead in the water without this software functioning. What's my next step? system restore point prior to 12.2 upgrade?
Hi! I have exactly the same issue, only since i've updated After Effect CC. Please help.
The first thing to do is trash your preferences folder....
1. go to a command prompt and type %appdata%
2. select the folder path Roaming/Adobe/After Effects
3. rename the 12.2 folder to anything else (example: 12.2old)
4. restart AE
If this still doesn't solve your problem, then...
5. google/download the app Adobe Cleaner - this is to wipe your system
registry and HDD of an app(s)
6. wipe AE from your system and then reinstall the app via CC Desktop app

what preferences folder are you talking about ? i can find it
i suspect on win10 its the appdata/roaming/adobe/aftereffects
but there is no preferences folder!?
