You should be able to use Live Paint, or you can select the line (make sure it is long enough) and Object>Divide Objects Below, to get two independent half circles to fill.

Larry G. Schneider Mar 25, 2014 12:04 PM (in response to BaCbDc)Draw the dividing line a little longer than the outer edge of the circle and use Pathfinder>Divide from the Pathfinder panel. It will trim the dividing line to the circle and give you two shapes to fill seperately.

Could be I'm making something wrong. But first solution worked for me.

One sure way of dividing accurately and safely is to use Smart Guides and the Line Segment Tool, ClickDragging from the top to the bottom Anchor Point of the circle (where Smart Guides say anchor).
In this case, Objects>Divide Objects Below with only the line selected and Pathfinder>Divide with both selected give the same result, apart from the latter having the two half circles form a Group, so you have to either Ungroup or Direct Select each half (or click each in the Layers palette) to colour them separately; the issue of one colour may be owing to that.

BaCbDc Mar 25, 2014 6:07 PM (in response to Larry G. Schneider)Now I managed it. I didn't know I have not only to mark the circle with the "Selection Tool"
but also mark the line with the "Selection Tool" in an extra step (hold shift to not unmark
the circle when I mark the line). When I marked only the circle the intersections of
the line with the circle were automatically marked as well. But as said I noticed
that was not enough. But is it possible to mark all paths automatically
instead to press Shift+"Selection Tool" for all paths separately?
Because if I have a lot of path like this it would be easier to mark all path at once:

As you have found, it is crucial to select all involved paths in connexion with Pathfinder (I mentioned that in post #5).
To select all, you may use Ctrl/Cmd+A if you have no other artwork, or you may ClickDrag with the Selection Tool making sure all paths are included, or you may ClickDrag with the Direct Selection Tool making sure all Anchor Points are included.

in your post no 5. But have now read Smart Guides are the help lines
of Illustrator and ClickDragging is what it says :-). And it works for me
all ways you described in post 7.

Depending on the settings you may see more or less, but basically Smart Guides are just helpful friends telling you what you are close to and guiding you to get there. I believe I ought to have written View>Smart Guides the first time.
