There is no proper way to do it. You either have to go into the ePub after export, or change the styling so there is a soft return between the chapter number and chapter title instead of a hard return. The second option will involve completly restyling, and getting the colon in there will be tricky.

This TOC issue has bugged me for years.
I have been able to come up with certain workarounds, but only when my physical (print) TOC just happens to be set up exactly as I would like it displayed in the ePub (this is not always the case).
It would be wonderful if there was a way for us to better control how the TOC info is output when exporting to ePub.
For now, the fastest and easiest way to deal with this issue (for me anyway), is to manually update it in the ePub file after export.
That's what we've been doing as well. Sigil has been our main ePub editor, but it is slowly getting buggier and buggier. Calibre, which we've used for various conversions and metadata control, actually has an editor now (which snuck up on us). There's some kinks to work out, but so far, we think it may be the Sigil replacement.
I use Oxygen.
It is still my favorite ePub editor.
