Are you sure you need the pixel aspect ratio to be 3,21:1 or do you just need the aspect ratio of the picture to be 3,21:1?
Please tell us the specifics of the project you're delivering: what are the specifications? How will your work be used next? How will it be played back? This is a VERY ODD pixel aspect ratio -- so odd that you could probably see the individual pixels in your finished render.

Situation looks like this:

I would think creating it in square pixels would produce a better final result, but that would depend on your playback hardware.
A 3840x1080 square-pixel comp sounds like a good starting point. But there is crucial information missing that prevents us from giving a definitive answer.
What are you expected to DELIVER? What are the specifications for the animations you are to create? How will your work be played back? All of these things are crucial in how you proceed, and we know none of them... so we can't help very much.
If your project is going to be displayed on two screens you need to know how it is going to be played back. If you are setting up synchronized playback of two video files set up your comp at 2X as wide as HD (3840 X 1080) but create two rendering comps that are standard HD and drag the 2X comp in each as a nested comp and adjust the position or anchor point to give you a left and right render.
If you are planning to setup the computer that plays back this file with two monitors then first you have to make sure that you can straddle the playback across both monitors. If so, then just render your 2X comp using a data rate that your media player will handle.
If you are not doing the playback then you MUST check with the client's equipment provider for specifications. Systems designed for playback on multiple screens do not necessarily follow any set standard you you must get the specifications from the supplier or your work product will only have a small chance of being usable.
Will you be using a digital signage system? Those have proprietary codecs, sometimes, and tight specifications. Be sure to ask Rick's questions of whoever you are supplying the movies to.
