You may:
1) Create one heart as a vector path;
2) Effect>Distort & Transform>Transform set the horizontal Move to the desired centre to centre distance between hearts and the desired number of copies (going right);
3) Repeat 2), only with a negative vertical Move (down);
4) Object>Expand Appearance;
5) Delete the unwanted hearts and recolour those to have other colours than the original.

Yes, if you don't mind that the fill is pixel based.
Follow Jacob's steps to create a grid of hearts on top of your image.
Make sure that each heart is the size of the enlarged "pixel". (If you do this enlargement in Photoshop, use the Nearest Neighbour to get hard edges for each "pixel").
Make sure that the Hearts grid is aligned with the large image Pixel grid.
Fill the Hearts with Black. Select them all and Group them.
Select both the Hearts and the picture below it and click Make Mask in the Transparency Panel. Check the Invert Mask option.
This should be the result (I added black background).
I just thought that it may be even better to convert the image fiirst to a vector mosaic: Object menu > Create Object Mosaic...
Then create the Heart grid and make it a Mask.
When you go to Object > Flatten Transparency and set the Raster/Vector slider to 100, you will get coloured heart shaped vectors

Sorry, I overlooked the fact that you were masking a raster image, so I was fooled by the image in post #5. In addition to the Opacity Mask suggested by Ton in post #8, you may also use the hearts as they are and (1) - 4) unchanged):
1) Create one heart as a vector path;
2) Effect>Distort & Transform>Transform set the horizontal Move to the desired centre to centre distance between hearts and the desired number of copies (going right);
3) Repeat 2), only with a negative vertical Move (down);
4) Object>Expand Appearance; this will give you a Group of hearts; you may Direct Select and delete some if desired
5) Object>Compound Mask>Make; you can do that because they are no overlaps;
6) Also select the underlying image and Object>Clipping Mask>Make; there is no need to change the colour of the hearts as with Opacity masks.
