Moved to InDesign epub forum.
I had a similar issue. The hyperlink in your document is no longer working.
If you go to Window -> Interactive -> Hyperlinks you will see your link with a red circle next to it. Just reenter the URL, and you'll be in good shape.
I have the same issue, but when I go to Window -> Interactive -> Hyperlinks I see my link with a green circle next to it, so that's not my problem.
Any other ideas?

Your issue seems to be tied to your user path. Check your library contents. You may have added a temp file in the "Cache" folder to your library and the "Cache" directory no longer has the data in it. I'm guessing you may have copy and pasted something into the library and the shortcut to it is no longer valid.
From your post:

so if you follow this path and it is empty it explains the error message.
When you test the resulting Epub file are any of it's links inoperable? If so that may point you to what is suppose to be in the /Caches sub-directory.
