The registry settings for autoplay did not get change after installing Lightroom 6 and that is why auto play is still trying to load Lightroom 5 on your machine and giving an error as it has been uninstalled.
The fix for this is to update the registry settings :
Press Windows + R keys to open run command and type regedit to open registry editor
and go to the path :
With the string data
This needs to be corrected (just remove 64) to:
I made the changes you suggested, but now nothing happens. When I attach the camera, Autoplay starts as usual. I select "Import with LR6", autoplay goes away and nothing happens.
If I start LR manually (double-click desktop shortcut) and look at Task Manager there is a Lightroom Process. If I try and invoke it via Autoplay and look at Task manager there is no lightroom process. My registry looks like:

you may try to update the following registry and do not forget to take the backup before updating the registry :
The original value might be:
C:\Program Files\Adobe\Adobe Photoshop Lightroom 5.7\Lightroom.exe "%L"
You need to change it to:
C:\Program Files\Adobe\Adobe Lightroom\lightroom.exe "%L"
Restart your machine after that and plug in you camera then.
Share the result after that.
OK, I've done as you suggested but there's still nothing happening. Autoplay goes away but the Lightroom process doesn't start. No difference from the previous attempt
PS. My previous attempt at this reply was rejected by a moderator. No reason given, nothing to suggest why it was rejected. I've edited it and resubmitted it but, not knowing why it was rejected, I have no idea whether this version will be accepted.
It is best NOT to use Autoplay for security reasons. Malware writers love Autoplay!!
I had never heard of that before, could you please give an idea of how this may manifest itself?
