You could do that with a track matte on an adjustment layer or you could use animated masks and add the mask to the effects using the compositing options in CC 2014 or LATER. You could also just blur some shape layers and mess with the blend modes and opacity to simulate the lighting effects. You could also use a layer with transparency as a gobo (matte) for a light if all the layers were 3D. It's all layers. You just have to play around until you get what you want. If you don't know about track mattes or Compositing options please use the Search Help field at the top right corner of AE to figure out how they work.
looks like you could recreate it using an animated gobo and a light on 3D layers. I've got a light shining through a matte made with a bunch of masks. Something like this:
But there are a zillion ways to make the same look.
if you want the movement like in stop motion. i usualy using posterize frame.
but you can use Rick's trick if you want the strobe light effect.
