I just pasted your code into a fresh Canvas document verbatim, drew a square and made it a button named nextBtn01, and the click event worked fine.
Try bringing up the developer console when you're testing the page (F12) and see if any errors are popping up.

I checked the developer console and it's throwing an error that says "TypeError: this.nextBtn01 is undefined" but I've double checked and the instance is named correctly. Thoughts?

Try actually copying and pasting the clip name from the code into the clip name field.
Try console.log(this.nextBtn01); If that just prints undefined, something's deeply awry.
It did return undefined, weird. It's just a simple keyframed animation too, strange.
I was able to cut and paste the pieces into a new FLA after getting the button working and it all seems fine now, no idea what the issue was.
