What do you mean it's not enabled in After Effects? Can you use the new GPU-enabled effects in After Effects CC 2015.3 like Lumetri or Gaussian blur?
My guess is that you are referring to the ray-traced renderer. Do you use that feature? I don't know of many people who do. There is a much more powerful way to create 3d geometry in AE now. However, if, for some reason, you want to use the ray-traced renderer, you can tick the box to "Use An 'Unsupported' GPU" (or however it's worded).
Other than the new GPU-enabled effects in CC 2015.3, here's what After Effects uses the GPU for. You'll notice it isn't much. Unless you are using the ray-traced renderer to create geometry, a GPU won't make your renders faster except with the three new GPU-accelerated effects in AE or certain third-party plugins.
Yes, some third-party plugins like Video Copilot's Element 3d, Red Giant Universe, Zaxwerks 3d Invigorator, Mettle's ShapeShifter, etc. do render on the GPU, but their plugins don't have anything to do with AE's ray-traced renderer or AE's GPU usage, so they should still be fine as long as your GPU is on the list for the specific plugin you plan to use.
