It sounds like you are used to working with Flash movie clips which have different button states in different frames. In Director, you need to use a behavior to swap the member of a sprite on mouseEnter, mouseDown and mouseLeave.
The Library Palette has a Rollover Change Member behavior. You can see this at work (with an animated filmloop for the rollover state) at http://nonlinear.openspark.com/tips/filmloop/rollover/.
You can also find a behavior which sets the states that you are looking for at http://nonlinear.openspark.com/tips/scripts/button/. Look for the Mouse States behavior.
Why do you want to work this way? If you want to avoid being misunderstood, it would help if you gave a clear explanation of what you are trying to do.
