Need help writing out average LAB values of color patches

I am trying to automate scanner settings, going through a large combinatorial matrix of parameters like Contrast, Brightness, Highlight, Shadow, Etc.

I have EZTWAIN set up with a Python script to iterate through the combinations.  I am scanning an MTF target, a GrayScale 15% Card and the GretaMacBeth 24 Color Chart.  I am trying to figure out how output each of the 24 LAB colors for the patches.  I figure if I give the coordinates of a 50x50 pixel square for each patch and take the average, I can see which setting combination has the least worst std dev from the Color Chart values.

All of the hundreds of scan combinations are in the same exact location, named with a prefix of each of the parameter/settings.

How would I open a directory of images/scans, check the 24 coordinate LAB colors and output the results to a .csv file with the file names?

I have used the PS jsx in the past to script all the plugins and their settings, but I haven't done anything requiring finding pixel values and writing out data.

Any help in the right direction would be appreciated.
