John, we'll need more information about your source media, the project settings you're using and the setup of your computer hardware. All these things are necessary to get an understanding of what might be the problem.
These issues tend to be because the person is using a different format source file to the project settings in Premiere and hasn't rendered the video.
Or... you're trying to playback footage that has a datarate higher than your hard drives can sustain.
Either way, more information is needed.

Need more info like, the format of the video footage, what codec, what is the bitrate per seconds, etc....
And, following on from Philip's message, what project settings you are using in Premiere. For instance, if you're trying to run HDV footage but you're only using a DV preset, there will be trouble.
