This is just After Effects telling you that what you're seeing on screen isn't the way it would look if it plays out in realtime.
If you get this when you're running a RAM Preview, wait for the RAM to clean and cache. It should "kick in" and play realtime.
If you get this when you press the spacebar to play the clip, it's because your machine is attempting to render from the HD on the fly, and it can't process as fast as your composition settings need it to in order to playback in realtime.
Try using RAM Preview
Indeed, as Ko said. If the problem persists even for RAM previews, then there's something going on. Could be anything from odd audio settings to problems with the disk cache. Check Ko's suggestions first and come back, if you need further assistance.
Make sure that the preview to external monitor is turned off in the Preferences (or with the Ctrl Numpad / shortcut) since AE can really be bogged down by 3rd party drivers that provide previewing to external monitors.
