There are two ways of doing this:
1. Using the minimum patch level tag in your application xml, so that it is equal to the current path of runtime available. http://help.adobe.com/en_US/AIR/1.5/devappshtml/WS5b3ccc516d4fbf351e63e3d118666ade46-7ff1. html
2. Or provide a badge for your application: http://www.adobe.com/devnet/air/articles/air_badge_install_02.html
For both of the above methods, runtime would be fetched first before the app install.

3). Express Installation
In these folder which one i need to use and how ? can you provide me a sample?
