The YouTube presets contained in the H.264 family of presets in the CS4 Adobe Media Encoder work exceptionally well for me from 720p Projects.
It is a very simple matter of using the bitrate to control the file size and quality in these.
I believe others ( maybe including Ann Bens if I recall from previous posts) find wmv to be excellent as well.
As Shooternz already pointed out: H.264 preset for YouTube widescreen HD.
With a slight tweeking. Those 5 minutes took a hell of a long time to render.
And my machine is not underpowered.
But yes i also like wmvHD 720p with the default settings.
Could be me but I think the wmv runs smoother then the mp4 on YouTube.
On Vimeo have not gotten a smooth playback yet.

Just checking on export- if your source was 1440x1080 50i and you output to 1920x 1080 did you also check the de interlace box?
Some people say Pr does a bad job at de interlacing (Harm) and you should let youtube do that?
Just wondering exactly what you did as results from 1440x1080 are amazing!!!
