I think that you're asking to be able to attach a layer to a joy stick then record the movement. If that is the case the answer is no.
If you want to create a motion path in real time you should look at Motion Sketch. Here's some info from the help files.
If your gaming joystick software allows you to emulate a mouse then you should be able to use the joystick and motion sketch.

If you can get the data from the joystick in any format outside of After Effects, then you should be able to create a script that converts that data into the format that After Effects uses. See this page for information about how you can copy and paste keyframe data from a spreadsheet or text file.
