You can create several things on Illustrator(+ other adobe products, specially fireworks and photoshop). I am not quite sure what kind of newsletter you want to create exactly. Do you have a link or some reference that you wish to imitate for your website and/or company?
Also, is this newsletter going to be in a website or you are going to print it?

No link or example--I simply want to create a 3 fold newsletter for print only. Artwork both sides. Paper size 8.5 x 14". I need to better understand if creating this in Illustrator is possible and the tools available to me to set up margins, column folds ect. Is there a tutorial which I can review and get me started on this project?
It can be done in Illy.
I believe there are templates for it, but you may design it yourself, and you may draft it using an actual 8.5 x 14, or in Illy.
I presume a letter fold (one panel inside the other two).
You may start by defining the folds. Remember the inside panel should be a bit narrower; the recommendation is some 1/32" to 1/8"; I would suggest something closer to the latter than to the former.
Then you may decide the widths of columns and margins (including those across the folds); with identical column widths, the side margin between the fold and column of the inside panel should be narrower.
And then you are just about there.
All this depends on the desired appearance.
