Thanks for your suggestion and thanks for looking into this... It works the way you suggested. Nevertheless after some troubleshooting I found out that you can do the single and multiple uploads without having to reload flash.
First, any time you start a new upload (either single or multiple) you have to create a new instance of the appropriate upload class (FileReference or FileReferenceList respectively). Then you have to make sure that when the upload(s) are completed you make that instance null. This removes any opened browsing sessions.
With the upload class instance made null next time you want to upload something, you are basically starting from scratch --and that's why you create a new upload instance with any new upload attempt.
This is only necessary if you're allowing users to do either multiple or single uploads simultaneously. If you are going to stick to a unique type of uploads ( single file only or multiple files only at any time) you can just declare the class instance once and don't need to make it null at the end of the upload process.

I am new to Flash development. I am also facing the same problem... I set the filereference object to null. but still it is not working, I have 2 mxml files where i am using filereference object. In one mxml it is working fine without any issue, where as it not working fine with second mxml file.

apply the cancel() method to your filereference instance.
Thanks for your reply... Issue is resolved using file=null; only the problem was browser was loading swf from cache. when I close browser and delete temp. files it works.
