You mean a new project in Premiere ? If so, match what the source material is....whatever your source is ( video characteristics ) is the settings for new project....make them the same....
Use the GSpot program or something like that to determine the source video characteristics
YouTube converts stuff to MP4 .....it will convert anything you upload to YouTube to MP4....and handles a lot of different formats that are uploaded OK.
It doesn't have to be FLV ( which is a Flash format ) It can be MP4 that you upload.
Once you figure out what your source video is and start a new project, that may determine the best format ( codec ) to use for exporting....
So, once you have that info and have got that far in your work flow....it would then be easier to help you with export settings..

Thanks for the info. My source is and AVI file which I export from Adobe Flash and can modify the size as needed. Does that information help? I am not sure waht settings to use when starting a new Premier Pro Project.

AVI is a format that can be any number of things " inside " the AVI file....kinda called a "wrapper"...GSpot will tell you the characteristics once you've got the AVI exported from flash ( I don't use flash so can't suggest settings on that end )...
For example, you might have an AVI file that is 720x480 progressive or interlaced , might be 29.97 fps or 23.97 fps...or 24, 30...lots of variables... since Flash is primarily used for web based apps....to show movies on digital computer screens.....and is not used for video on TV ....I have no idea what characteristics your AVI file will have...
Gspot will help you discover that....it's a free program....
Then your project in Premiere should match what that source video is....same size, frame rate, etc.....

I have a movie I made in Flash which I can export as an avi. I want to import that into premiere and export out of premiere for Youtube. I hope this helps.

If this is in flash, just export it as an flv.
Second, even though YouTube plays flash movies, flash is not usually the best upload. I use vimeo, so every time I use YouTube, I look at their latest recommendations for formats, datarates, etc to upload.
Related to your earlier comment about resizing, for a flash, I'd consider a high def size to take advantage of that option.
