If that doesn't help, try resetting your PS preferences as described in the FAQ.

You either have to physically delete (or rename) the preference files or, if using the Alt, Ctrl, and Shift method, be sure that you get a confirmation dialog.
This resets all settings in Photoshop to factory defaults.
A complete uninstall/re-install will not affect the preferences and a corrupt file there may be causing the problem.

Glad it fixed it.
When the preferences get corrupted it is not due to a wrong setting. One of the files has been damagrd. This is a known problem with computers not just Photoshop.
It can be caused by incorrect shut-downs, hard drive faults, or a dozen other reasons. Deleting the preferences just lets Photoshop generate a fresh set at factory default.
Make sure you check through to get your personal settings back and then save a copy.

Jack Oct 27, 2010 2:12 PM (in response to John Joslin)Problem has returned on second attempt to use clone stamp tool. Tried reopening with Ctrl+Alt+shift depressed, got the "delete preferences file" dialog. Still unable to use clone stamp tool on flat file or background layer of layered image.
Further ideas appreciated.

Bad news is that a stop/start with reset key combination is required after every instance of using my clone stamp tool.
Any ideas of what can be the cause, and a possible cure, would be appreciated.

I would try these simple to do checks on your computer to help rule out problems.
Go to start and type in CMD. Your will see CMD.exe. Right click on that and choose run as Admin. Type in scf /scannow at the command prompt. This will check windows 7 system files and if problems are found will fix them. It may ask you for the disk or to reboot. Keep doing this until nothing is found. If nothing is found then everything is ok.
You should also scan the hard drive for disk errors. Something might be on a bad sector or trying to write to a bad sector.
