A black or dark layer with the signature as a transparency mask should work. Assuming you have the signature as dark on a white background.
1. Use the rectangular marquee and copy the signature.
2. File - New to create a new image the size of the copied signature (Clipboard preset).
3. Layer - New Fill Layer - Solid Color, make it Black.
4. The layer will be given a layer mask by default.
5. In the Channels palette, make the Color Fill 1 Mask channel current.
6. Paste in your signature.
7. Image - Adjustments - Invert the colors (so black is white and white is black).
8. Delete the background layer.
9. Save as a 24 bit PNG.
You should be able to come up with something like this partially transparent PNG:
