Well, one possible reason could be that you are working from a networked drive. Where is your Project located? Where are the Scratch Disks directed?
Another possibility could be something like a USB external. Do you have any external HDD's in your system? If so, what is the connection type?
Yet another could be a lack of defragmented free space (though PRPROJ files are usually not THAT large, unless you have been bitten by Project File Bloat - if so, update to 5.0.3). Do you have adequate, defragmented free space?

Start by providing DETAILS about... everything
Beginning with your Windows version and your user level
Also, right click the program icon and select the Run As Administrator option to be sure the program has full permissions

After 30 minutes, it saved and closed the program. And upon reopen
ing, everything is there. I am in the process of defraging the scratch drive disk...

I cleaned up the scratch disk hard drive and defraged it and the auto save and manual save is working again.
