After Effects woould be the better tool to use.

If you're trying to correct using AE, check out Video Copilot's basic AE tutorials (http://www.videocopilot.net/basic/#01). I'm sure there'll be something there that'll help (that's how I moved forward from zero on AE).
If you do a search on the Adobe TV site you should find a tutorial by Karl Soule of 'Short and Suite'
which shows you how to use Photoshop to alter the type of blemish to which you refer
Was done under CS4 but can't imagine there will be any major differences in your case.
