This is an issue with registry permissions.
Do you try to install the update as an Administrator user?
What is your Windows version?
What Reader version are you trying to update?
I log in Windows using Admin account.
My Adobe Reader updates by itself automatically. Everytime it will notice me by "Are you trying to install these updates?" etc and I click "Yes". Then it will install the updates that have been downloaded automatically.
I am using "Windows XP Professional, version 2002, Service Pack 3".
My Adobe Reader is "Adobe Reader X, version 10.0.0".
You can try to manually download and install the update from ftp://ftp.adobe.com/pub/adobe/reader/win/10.x/10.0.1/misc/ - see http://kb2.adobe.com/cps/888/cpsid_88814.html#main_Tiers what to download for your Reader version.
But it is quite possible that you may get the same error while trying to install the update. In this case you should check the permission settings in the registry for key HKLM\SOFTWARE\Microsoft\Windows NT\CurrentVersion\Image File Execution Options\AcroRd32Info.exe
