I am looking for advise on an issue with live corners that do work well for objects with 4 corners.
Now I drew a stylized S-character with the pen tool and would like some of the corners to be
round - some of them straight.
As the corner option dialog box only works for rectangles, the chain icon is greyed out in my case
and therefore I am not able to change a specific corner that way - any suggestions or workarounds?

If you're creating custom paths with the Pen tool, you'll need to use the path editing tools (Pen tool, Direct Selection tool, Convert Direction point tool, etc.) to create the corner you like. As you found out, the Corner Options only work with corners at a 90-degree angle.
There's also a CornerEffects sample script that will work on any shape that has some potential to help you, but I'm not certain it will work for your case. In the script, the points are counted sarting with where you click the pen first, then working counter-clockwise to each new click.
