If I try to learn how to use HTML5 for my video and audio clips instead of using Flash, does it mean that at the moment my clips won't show in some browsers?
Not necessarily because there are some HTML tags that are backwards compatible and to insert a video or audio clips, you can use the normal HTML tags to do exactly what you want to do.
I would strongly recommend you to start learning HTML5 and CSS3 because all the new browsers are supporting them and people are using new browsers because they want to continue using new features of youtube, yahoo, hotmail, facebook and twitter who are developing their sites based on HTML5/CSS3.
You best bet is to do something in HTML5/CSS3 and post your page here for people to comment. Dreamweaver forum is better for this because there are more people using it.

I'm using Dreamweaver CS4, I intend to upgrade when CS6 comes out.

Sure you can start now by downloading a trial version of DW CS 5.5 and use it for 30 days. When this expires, you can then download Microsoft's Expression Web 4 and use that for another 30 days. By then in the new year we'll have CS 6.0 and you can buy it to continue learning with it.
