Did you switch cables recently? I have 3 USB cables but only one of them works well. Now I use an amplified cable if I need any length more than 1.5 meters.
I don't know what to suggest other than that. I myself stopped using LR to tether a 5D2 with a Mac because, to put it crudely, it's crap with absolutely no consistency.
Now I use C1 Pro with no issues whatsoever, it never fails me and no embarrassing annoyances anymore.

I say this because with my Macbook Pro i noticed on very rare occasions the USB port would fail until i restarted the computer which seemed to juice it up again, if you haven't done a complete shutdown try that. Alternatively test something else in the same port like a printer or phone.
You could load the software called disk utility from canon and try and tether in canon's DPP software.. It should have come on a disc with your camera. If that works then at least we know for sure the problem is with LR.

This actually did help me. I tried to Image Capture and Lightroom on two different macs, with three different cords. Finally, I went into apple>about this mac>more info> USB and the camera did not show up on either computer. I called Cannon and it seems the port on the camera where the usb plugs in is defective, so I need to send the camera in for repair.
Sigh.

Simon Full Dec 30, 2011 11:03 AM (in response to SF Ellen)You can buy special clip cables that take away the stress from the USB entering the camera by creating a small loop. These are a really good idea as you probably pulled or bent the connection at some point, easy to do.

This is fantastic! I realized too that right before the tethered shooting stopped functioning, I tripped over the cord--not enough to pull anything over but the cord was jerked out of the camera--I am sure that this is when the port became damaged.

Simon Full Jan 16, 2012 8:05 PM (in response to monmart)There is no tricks.. it's a USB2 bottleneck for sure as lightroom is only a tad slower than C1.. Cam manafactures need to give us USB3 or Thunderbolt but i don't see that happerning anytime soon. I wish USB2 like Flash would just up and die already.
